The samba cell is the one most people mean when they say a cavaquinho "plays samba": down and up strokes alternate through the bar while a mute chops in on every off-beat, four mutes in eight slots — the busiest chop count of the three cells. The mutes are load-bearing: they are what makes the down and up strokes around them read as a groove instead of a scale exercise.

The pattern, step by step

Getting it to sit

Isolate the mutes before you trust yourself with the strokes around them. The commonest fault in this cell is letting a mute ring a little — the fretting hand lifts just enough for a ghost pitch to leak through, and the chop stops sounding percussive. Damp fully, strike cleanly, and only then bring the down and up strokes back in around it. When the four mutes sound like four identical clicks, the cell is starting to work.

Then loop it under a chord you can hold without thinking, so the whole exercise is the right hand. A samba spends whole choruses on the same short chord loop, so the chop comes around every bar all night; run it against the metronome until the mutes land on the beat without you counting them.
